Abstract
We have developed a method that performs RNA extraction, size selection, and purification in a single microfluidic procedure. We leverage an electric-field based focusing method called isotachophoresis (ITP) and on-chip polyacrylamide gel electrophoresis to overcome the substantial sample loss issues existing in conventional methods. We demonstrate the high resolution and high yield of our method by purifying RNA molecules in the range of 20 - 35 nt with a yield of 80%. Our method is compatible with downstream assays and can be easily customized for different size ranges, which potentially enables sample preparation required for sequencing RNA of specific size from extremely low abundance samples.
